HCC1806 Cells
Basic details about HCC1806 cells
Description | The HCC1806 cell line is derived from the mammary gland of a 60-year-old patient with acantholytic squamous cell carcinoma. These cells lack receptors for estrogen and progesterone, and the absence of epidermal growth factor receptor (EGFR) amplification, categorize it as a triple-negative breast cancer. The cell line is instrumental for the biological validation of therapeutic targets, as it closely mirrors the behavior of TNBC in vivo, including tendencies for spontaneous metastasis and resistance to conventional therapies like paclitaxel. Molecular effects of interventions, such as AEB071 treatment, on HCC1806 cells, provide insights into the cell proliferation pathways and the potential of protein kinase inhibitors as therapeutic agents. The use of HCC1806 in xenograft models contributes to the study of tumor growth and metastasis in a controlled environment. HCC1806 breast cancer cells serve as a valuable tool for the study of breast cancer, particularly within the context of triple-negative subtypes. It stands as a critical resource for researchers looking to unravel the molecular interactions in breast cancer and search for effective treatments against this challenging variant of the disease. |

Cell culturing methods
Culture Medium | RPMI 1640, w: 2.1 mM stable Glutamine, w: 2.0 g/L NaHCO3 (Cytion article number 820700a) |
---|---|
Medium supplements | Supplement the medium with 10% FBS |
Passaging solution | Accutase |
Subculturing | Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium. |

Quality control of HCC1806 cells
Sterility | Mycoplasma contamination is excluded using both PCR-based assays and luminescence-based mycoplasma detection methods. To ensure there is no bacterial, fungal, or yeast contamination, cell cultures are subjected to daily visual inspections. |
