| 說明 | The UM-UC-3 cell line is derived from a human bladder carcinoma, specifically a high-grade transitional cell carcinoma (TCC), established from a male patient. It has been widely used in cancer research due to its robust growth characteristics, both in vitro and in vivo. UM-UC-3 cells display an epithelial morphology and are aneuploid, with a modal chromosome number ranging from 59 to 95. These cells are capable of forming tumors in immunocompromised mice, with histological features resembling the primary tumor, highlighting their utility as a preclinical model for bladder cancer. Genetic and molecular studies have revealed significant alterations in UM-UC-3 cells, including frequent deletions and mutations in key tumor suppressor genes such as CDKN2A and CDKN2B. These genes are located in the 9p21 region, which is commonly deleted in bladder cancer, contributing to dysregulation of the cell cycle. Additionally, UM-UC-3 exhibits changes in the phosphatidylinositol 3-kinase (PI3K) signaling pathway, a critical driver of tumorigenesis in urothelial carcinoma. | 傳代培養 | Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium. |
