| Descrição | HROG59 is a primary human glioblastoma multiforme (GBM) cell line established from freshly resected tumor tissue of an adult patient diagnosed with WHO grade IV glioblastoma. The cell line was generated as part of a systematic effort to develop ultra-low passage glioma models preserving patient-specific biological and molecular characteristics. HROG59 cells grow adherently in vitro and display a fibroblast-like morphology typical of primary GBM cultures. The line has been maintained under standardized culture conditions to minimize genetic drift and maintain phenotypic stability. HROG59 has been used to evaluate sensitivity to standard-of-care and investigational agents in glioblastoma therapy, including alkylating agents and targeted inhibitors. As a low-passage, patient-derived GBM model, HROG59 provides a clinically relevant in vitro system for studying glioblastoma biology, intertumoral heterogeneity, and therapeutic response mechanisms, while maintaining close fidelity to the original tumor specimen. |

| Subcultura | Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium. |
