| Description | HCC1937 is a human breast carcinoma cell line derived from a primary tumor of an adult female. This cell line exhibits several genetic alterations characteristic of aggressive breast cancer phenotypes, including a homozygous mutation in the BRCA1 gene (5382C mutation), which is a notable marker for predisposition to breast cancer. The presence of this mutation aligns with a familial pattern of breast cancer as it is also detected in other family members, indicating a hereditary aspect to the malignancy. Additionally, HCC1937 has an acquired mutation in the TP53 gene coupled with the loss of the wild-type allele, further compounding its tumor suppressor deficiencies. The cell line also displays a homozygous deletion of the PTEN gene and exhibits loss of heterozygosity at multiple loci involved in cancer pathogenesis, suggesting a complex genetic background conducive to oncogenic transformation. From a phenotypic perspective, HCC1937 does not express the estrogen receptor (ER) or progesterone receptor (PR), categorizing it as ER-negative and PR-negative, which are typical markers for more aggressive disease courses. | Repiquage | Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium. |

| Conditions d'entreposage | For long-term preservation, place vials in vapor-phase liquid nitrogen at about −150 to −196 °C. Storage at −80 °C is acceptable only as a short interim step before transfer to liquid nitrogen. |
