CHO-NECTIN4 cells are a stable recombinant Chinese hamster ovary (CHO) cell line engineered to express human Nectin-4 (also known as PVRL4 or Poliovirus receptor-related protein 4), a type I transmembrane protein belonging to the nectin family of cell adhesion molecules. Nectin-4 is a well-established tumor-associated antigen that is overexpressed in multiple solid tumor types, including urothelial bladder carcinoma, breast cancer, non-small cell lung cancer, and pancreatic cancer, making it a clinically validated target for antibody-drug conjugates (ADCs) and other targeted immunotherapeutics. The ADC enfortumab vedotin, which targets Nectin-4, is approved for treatment of urothelial carcinoma, underscoring the therapeutic relevance of this antigen. CHO-NECTIN4 cells are widely used for the development and characterization of Nectin-4-directed antibodies, ADCs, bispecific antibodies, and CAR-T cell therapies. The stable recombinant expression system supports quantitative binding assays, ADCC/CDC cytotoxicity evaluations, receptor internalization studies, and high-throughput antibody screening by flow cytometry. The CHO background provides low endogenous expression of most human surface antigens, ensuring that observed signals are attributable to the stably expressed Nectin-4 transgene. This cell line is validated for use in drug discovery workflows, preclinical candidate selection, and mechanistic studies of Nectin-4 receptor biology. |

| Repiquage | For routine adherent cell culture: Aspirate the old culture medium from the adherent cells, and wash them with PBS to remove any remaining medium. After aspirating the PBS, add the appropriate volume of Trypsin/EDTA solution based on the culture vessel size (e.g., 1 ml for a T25 flask, 3 ml for a T75 flask) and incubate at room temperature or 37°C for 5-10 minutes, or until the cells detach. Monitor detachment under a microscope, and gently tap the vessel if necessary to release the cells. Once detached, add complete medium to inactivate the Trypsin/EDTA, gently resuspend the cells, and transfer an aliquot of the cell suspension into a new culture vessel containing fresh medium. Place the vessel in an incubator set to 37°C with 5% CO2, and change the medium every 2-3 days. |
