General information
| Description | HCT-8 cells, also known as human ileocecal colorectal adenocarcinoma cells, are an epithelial cell line originally derived from a 67-year-old Caucasian male patient with ileocecal adenocarcinoma. The HCT-8 cell line was established in the late 1960s and is widely utilized in cancer research, particularly in the study of colorectal cancer pathogenesis, metastasis, and treatment response. Morphologically, HCT-8 cells are epithelial-like and exhibit a monolayer growth pattern with a polygonal shape. They possess the capability to grow in both adherent and semi-suspended cultures, which is characteristic of some transitional stages of cancer cell metastasis. This feature makes them particularly useful for studies related to cancer cell invasion and migration. Genotypically, HCT-8 cells are hypertriploid, containing several chromosomal aberrations common in colorectal carcinomas, including mutations and deletions which are relevant to cancer progression and resistance mechanisms. This genetic profile supports their use in oncological studies, especially those focusing on genetic pathways involved in tumorigenesis and drug resistance. Research utilizing HCT-8 cells has contributed significantly to the understanding of colorectal cancer biology, including the elucidation of molecular pathways involved in cancer cell proliferation, apoptosis, and chemoresistance. | Subculturing | Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium. |

| Storage Conditions | For long-term preservation, place vials in vapor-phase liquid nitrogen at about −150 to −196 °C. Storage at −80 °C is acceptable only as a short interim step before transfer to liquid nitrogen. |

| Sterility | Mycoplasma contamination is excluded using both PCR-based assays and luminescence-based mycoplasma detection methods. To ensure there is no bacterial, fungal, or yeast contamination, cell cultures are subjected to daily visual inspections. |
